Abstract: | In a study conducted during 1998-2000 in China, the soyabean mosaic virus (SMV)-resistant soyabean line 95-5383 was crossed with 4 susceptible cultivars/lines (HB1, Tiefeng21, Amsoy and Williams) and one resistant cultivar (PI486355). Their F1 and F2 hybrids were then evaluated for SMV resistance by inoculation with SMV3. In the four resistant x susceptible crosses, F1 hybrids were susceptible while F2 hybrids exhibited a 1 resistant: 3 susceptible ratio, indicating that line 95-5383 carries one recessive gene that confers SMV resistance. A segregation of susceptibility was observed in the F2 progenies from the cross 95-5383 x PI486355, indicating that the SMV resistance genes in both parents are located at different loci. By bulked segregant analysis of F2 populations from the cross 95-5383 x HB1, one co-dominant RAPD marker (OPN11980/1070) closely linked to the SMV resistance gene, amplified with the RAPD primer OPN11, was identified. The DNA fragment OPN11980 was amplified in the resistant parent 95-5383 and resistant bulk, while OPN111070 was amplified in the susceptible parent HB1 and the susceptible bulk. OPN11980/1070 was amplified in the F1 progenies. Identification of the markers in F2 plants showed that the co-dominant marker OPN11980/1070 is closely linked to the SMV resistance gene in 95-5383, with a genetic distance of 2.1 cM. |
